The present thesis seeks to further understanding of luteal maintenance and regression by characterizing the hormonal environment and luteal transcriptome during these processes over the course of four studies. The first characterized circulating prostaglandin F2 alpha metabolite (PGFM) and progesterone (P4) during pregnancy and regression, observing minimal secretion of PGFM in early pregnancy, likely contributing to luteal maintenance at this time. However, in the second month of pregnancy distinct pulses of PGFM were observed, suggesting a different mechanism of luteal maintenance after the maternal recognition of pregnancy period. In animals undergoing regression, the classic pulsatile pattern of PGF secretion was observed, though the majority of decline in P4 was associated with a single large pulse, with no change associated with early pulses of PGF. However, focusing on these early pulses in the second study, there were 210 genes altered as a result of these early pulses within the corpus luteum, with the majority of these novel to the process of regression. Pathways altered in early regression were not predicted to alter luteal function, but rather decreased cell survival pathways, potentially increasing tissue susceptibility to later pulses of PGF. The third study characterized the luteal transcriptome in pregnancy, with 94 altered transcripts observed in the first month, with nearly 20% regulated by interferons. Surprisingly, a substantial subset of these remained elevated into the second month of pregnancy, though IFNT secretion is minimal at this time. Few genes associated with luteolysis were altered in pregnancy, indicating luteal maintenance is not transcriptomically the opposite of regression. Finally, in the fourth study, stable genes were identified across all physiologic states considered in the previous studies, with clear differences noted between groups in overall gene stability. From a physiologic view, this highlighted differences in gene stability in the CL during regression and pregnancy, while practically improving future selection of reference genes for RT-qPCR.
